Amino acid sequencing and sequence homology analyses of human melanin-concentrating hormone (MCH) from porcine and human sources show extensive amino acid sequence identity. We report the structure of the human MCH tetrapeptide Cys-Tyr-Tyr-Arg-NH2 based on X-ray diffraction analysis of a model molecule. The model for the MCH peptide was constructed based on the elucidated structure of the porcine MCH tetrapeptide. The crystal structure of the human MCH peptide was determined using multiple isomorphous replacement at 1.8 A resolution. The MCH model consists of three antiparallel beta-pleated sheets connected by a characteristic turn. The position of the tetrapeptide sequence is stabilized by extensive network of hydrogen bonds between the main chain atoms. These findings suggest the peptide moiety of MCH binds in the same site as the neuropeptide Y.Polymerization of cyclic di- and trinucleotides.
The first in situ studies of the polymerization of di- and trinucleotides are presented. We have found that while di- and trinucleotides polymerize in an identical fashion, their rate and magnitude of polymerization is different. (A)2A2 is a substrate for the polymerization of A3; the kinetics are very slow, but increase with the concentration of A3.
